Safety Considerations

  • Conditions linked to contraindication or caution (knowledge graph, not label text): anuria, hypercalcaemia, nephrocalcinosis, phosphorus metabolism disease, impaired renal function, intracranial haemorrhage, osteonecrosis of the jaw, hypophosphataemia, hypoparathyroidism, pulmonary oedema, fibromyalgia, myositis, tooth disease.
  • Drug interactions: 100 interactions were found. Moderate-level examples include amphotericin B (including lipid, cholesteryl sulfate and liposomal forms), aminoglycosides (kanamycin, neomycin, streptomycin), vancomycin, polymyxin B, aspirin, mesalazine, balsalazide, olsalazine, sulfasalazine and exenatide. Several interact through additive renal or electrolyte effects. Interactions with unknown severity include calcitriol, vitamin A, pantoprazole, glimepiride and doxycycline.
  • Calcium and vitamin D: Published case reports describe prolonged or severe hypocalcaemia after zoledronic acid, particularly with vitamin D deficiency. Vitamin D and calcium status should be corrected before use.
